Organic solvent- and detergent-resistant proteases are important from an industrial viewpoint. However, they have been less frequently reported and only few of them are from actinomycetes. A metalloprotease from Streptomyces olivochromogenes (SOMP) was purified by ion exchange with Poros HQ and gel filtration with Sepharose CL-6B. Apparent molecular mass of the enzyme was estimated to be 51 kDa by sodium dodecyl sulfate–polyacrylamide gel electrophoresis and gelatin zymography. The activity was optimum at pH 7.5 and 50 °C and stable between pH 7.0 and 10.0. SOMP was stable below 45 °C and Ca2+ increased its thermostability. Ca2+ enhanced while Co2+, Cu2+, Zn2+, Mn2+, and Fe2+ inhibited the activity. Ethylenediaminetetraacetic acid and ethylene glycol-bis (β-aminoethyl ether)-N,N,N′,N′-tetraacetic acid, but not phenylmethylsulfonyl fluoride, aprotinin, and pefabloc SC, significantly suppressed the activity, suggesting that it might be a metalloprotease. Importantly, it is highly resistant against various detergents, organic solvents, and oxidizing agents, and the activity is enhanced by H2O2. The enzyme could be a novel protease based on its origin and peculiar biochemical properties. It may be useful in biotechnological applications especially for organic solvent-based enzymatic synthesis.  相似文献

We have synthesized two cyclopentadithiophene (CDT)‐based low bandgap copolymers, poly[(4,4‐bis(2‐ethyl‐hexyl)‐4H‐cyclopenta[2,1‐b:3,4‐b′]dithiophene‐2,6‐diyl)‐alt‐(benzo[c][1,2,5]selenadiazole‐4,7‐diyl)] (PCBSe) and poly[(4,4‐bis(2‐ethyl‐hexyl)‐4H‐cyclopenta[2,1‐b:3,4‐b′]dithiophene‐2,6‐diyl)‐alt‐(4,7‐dithiophen‐2‐yl‐benzo[c][1,2,5]selenadiazole‐5,5′‐diyl)] (PCT2BSe), for use in photovoltaic applications. Through the internal charge transfer interaction between the electron‐donating CDT unit and the electron‐accepting benzoselenadiazole, we realized exceedingly low bandgap polymers with bandgaps of 1.37–1.46 eV. The UV–vis absorption maxima of PCT2BSe were subjected to larger hypsochromic shifts than those of PCBSe, because of the distorted electron donor–acceptor (D–A) structures of the PCT2BSe backbone. These results were supported by the calculations of the D–A complex using the ab initio Hartree‐Fock method with a split‐valence 6‐31G* basis set. However, PCT2BSe exhibited a better molar absorption coefficient in the visible region, which can lead to more efficient absorption of sunlight. As a result, PCT2BSe blended with [6,6]‐phenyl‐C61‐butyric acid methyl ester (PC61BM) exhibited a better photovoltaic performance than PCBSe because of the larger spectral overlap integral with respect to the solar spectrum. Furthermore, when the polymers were blended with PC71BM, PCT2BSe showed the best performance, with an open circuit voltage of 0.55 V, a short‐circuit current of 6.63 mA/cm2, and a power conversion efficiency of 1.34% under air mass 1.5 global illumination conditions. © 2010 Wiley Periodicals, Inc. IT capability as an important source of competitive advantage has been strongly emphasized in the strategic management literature, yet the formation and evolution of IT capability in the ever-changing business environment are not well explained. To fill this gap, this paper takes routine as the unit of analysis, depicts a micro-interpretation of the formation and evolution of IT capabilities from the routine-based and dynamic perspective, and uses the agent-based simulation methodology to simulate the evolutionary process of IT capability as well as to identify the underlying principles. In order to provide a better presentation of the evolutionary process, a routine-based view of the enterprise explicitly recognizes relationships of IT resources and capabilities. The simulation results show that the evolution of IT capability is a dynamic adaption and learning process. From the routine-based view, the evolution of IT capability is indeed the process of variation, selection, and retention for IT routines.  相似文献

A new cyclization route, triggered by epoxide opening, has been performed to provide the key intermediates for isoindolobenzapine alkaloids, lennoxamine and chilenine. The epoxide was prepared by the Stille reaction using vinyltributylstannane and the following dioxirane treatment. Cyclization under the treatment of BF3 · OEt2 provided an azepine moiety, and the oxidative cyclization toward the known precursor for the alkaloids has been achieved by reaction with a stoichiometric amount of Pd(OAc)2. This formal synthesis suggests a new route to the alkaloids.  相似文献

A multi‐channel microchip electrophoresis using a programmed step electric field strength (PSEFS) method was investigated for fast parallel detection of feline panleukopenia virus (FPV) DNA. An expanded laser beam, a 10× objective lens, and a charge‐coupled device camera were used to simultaneously detect the separations in three parallel channels using laser‐induced fluorescence detection. The parallel separations of a 100‐bp DNA ladder were demonstrated on the system using a sieving gel matrix of 0.5% poly(ethylene oxide) (Mr = 8 000 000) in the individual channels. In addition, the PSEFS method was also applied for faster DNA separation without loss of resolving power. A DNA size marker, FPV DNA sample, and a negative control were simultaneously analyzed with single‐run and one‐step detection. The FPV DNA was clearly distinguished within 30 s, which was more than 100 times faster than with conventional slab gel electrophoresis. The proposed multi‐channel microchip electrophoresis with PSEFS was demonstrated to be a simple and powerful diagnostic method to analyze multiple disease‐related DNA fragments in parallel with high speed, throughput, and accuracy.  相似文献

During the process development for multistep synthesis of Rosuvastatin calcium several impurities were obtained along with the final Rosuvastatin calcium. Out of this; synthesis of impurity A (acetone adduct) a minor impurity of Rosuvastatin calcium (3R,5S,6E)-7-[4-(4-fluorophenyl)-2-[[(2-hydroxy-2-methylpropyl)sulfonyl(methyl)amino]-6-(1-methylethyl)-pyrimidin-5-yl]-3,5-dihydroxyheptenoicacid hemicalcium salt, is described. The synthesis of impurity A has been accomplished in 6 steps; starting from formation of β-hydroxy sulfonamide as the key intermediate and followed by using convenient routes with overall yield of 13.5%. The target compound can be used as the reference substance of impurity of the Rosuvastatin calcium.  相似文献

We study two types of dynamical extensions of Lucas sequences and give elliptic solutions for them. The first type concerns a level-dependent (or discrete time-dependent) version involving commuting variables. We show that a nice solution for this system is given by elliptic numbers. The second type involves a non-commutative version of Lucas sequences which defines the non-commutative (or abstract) Fibonacci polynomials introduced by Johann Cigler. If the non-commuting variables are specialized to be elliptic-commuting variables the abstract Fibonacci polynomials become non-commutative elliptic Fibonacci polynomials. Some properties we derive for these include their explicit expansion in terms of normalized monomials and a non-commutative elliptic Euler–Cassini identity.  相似文献

This study investigates the effects of intermediate temperature heat release (ITHR) on autoignition reactivity of full boiling range gasolines with different octane sensitivity through intake temperature and simulated exhaust gas recirculation (EGR) sweeps in a homogenous charge compression ignition (HCCI) engine. To isolate the ITHR effects, low temperature reactivity was suppressed through the use of high intake temperature and low intake oxygen mole fraction. For quantification of ITHR, a new method was applied to the engine data by examining the maximum value of the second derivative of heat release rate. Combustion phasing comparisons of fuels with octane sensitivity showed that fuel with less octane sensitivity became more reactive as intake temperature and simulated EGR ratio decreased, while fuel with higher octane sensitivity had a reverse trend. For all of the fuels that were tested, the amount of ITHR increased as the intake temperature and oxygen mole fraction increased. These ITHR trends, depending on octane sensitivity, were almost identical with the trends of combustion phasing, showing that ITHR significantly affects fuel autoignition reactivity and determines octane sensitivity.  相似文献
